Dr. Jeffrey Stansbury and a multi-disciplinary team received one of the nine Anschutz Acceleration Initiative (AAI) grants. Multimaterial 3D print processing of antimicrobial and antifungal dental prosthetics. Year 1 funding totals $1,072,225.

Investigating the Role of Heterotrimeric G Proteins in Craniofacial Development and Auriculocondylar Syndrome

Pattern formation of the pharyngeal arch establishes the identity of cells that give rise to the bone, cartilage, and connective tissue of the face. Disruptions to this process can lead to a variety of craniofacial birth defects. The overarching goal of this project is to elucidate cell signaling pathways that establish cellular identity so that we can better understand mechanisms of development and disease.

Characterization of PDGFR alpha/beta heterodimers

Platelet-derived growth factor (PDGF) signaling has been shown to regulate numerous processes throughout the body, both during development and adult life, and aberrant PDGF signaling has been causally associated with defects in craniofacial development, cancer, vascular disorders and fibrotic diseases. Defects in craniofacial development comprise one of the most prevalent birth defects in humans, with an estimated incidence in the United States of 1/940 live births for cleft lip and 1/1,574 live births for cleft palate. The studies proposed here will provide valuable insight into the mechanisms underlying craniofacial development by characterizing dimerization, activation, trafficking and signal molecule binding dynamics for PDGFR a/β heterodimers, which may contribute to the development of new therapeutic directions for the treatment of human craniofacial birth defects.

Light-propelled dental adhesives with enhanced bonding capability

The study explores mechanisms by which dental adhesives can be given ‘deep roots’ within dentin, thereby strengthening the overall composite restoration. The adhesive layer within composite restorations (the so-called tooth-colored restorations) is often called the ‘weakest link’ as they are the first layer to fail, causing patient discomfort and the need for additional dental work. The smart nanogels developed in the Nair Lab can be dispersed within commercial dental adhesive formulations such that they swell up and ‘walk away’ from the surface, thereby driving the adhesive deeper within the dentinal tubules.  Such well-anchored adhesives have the potential to strengthen composites, leading to improved function and longevity of such restorations. This work is a collaboration with the Stansbury lab group.

Uniquely high conversion and mechanically robust composite restorative materials for functionally elevated performance

Resins that form the matrix phase of dental composite restorations are typically based on monomers such as BisGMA and UDMA that engage in hydrogen bonding, which serves to reinforce polymer networks. The focus of this project involves a redesign of the number and placement of urethane groups within one of the monomers in combination with complementary diluent comonomers that provide a strong noncovalent engagement with the urethane groups. The result is composite photopolymers that display extremely high strength, hardness and toughness. Additional work will explore the structure-property relationships that include assessment of reactivity, optical properties, wear resistance, fracture toughness and others to allow us to advance excellent candidate materials as potential new dental restorative materials.

Mechanisms patterning the midface

The human facial skeleton originates from groups of cells in specific positions with unique identities, each instructing the formation of a different skeletal structure. We hypothesize that in human genetic diseases, like frontonasal dysplasia, the ability to establish these identities is disrupted. In this proposal we use zebrafish disease models to investigate a hypothetical identity ‘code’ instructing positional cellular identity, thus informing the etiology of these devastating birth defects.

The ROS1 -ders, Inc. $75,000
Novel murine ROS1+ cancer models for next-generation therapeutics

ROS1 gene rearrangements encoding oncogenic fusion proteins occur in 1-2% of lung adenocarcinomas as well as subsets of glioblastomas, cholangiocarcinomas, Spitzoid neoplasms and inflammatory myofibroblastic tumors. Precision oncology with ROS1-targeting tyrosine kinase inhibitors (TKIs) including crizotinib and entrectinib has greatly improved outcome in ROS1+ lung cancer patients, but as single agents, they rarely provide cures. This project will develop genetically-engineered mouse models (GEMMs) of ROS1+ lung cancer and implantable murine lung cancer cell lines to pursue studies that may overcome multiple hurdles to achieving long-term therapeutic management or cures in these molecularly-defined cancers.

Evaluation of School-Based Health Centers (SBHC) in Colorado

The study will evaluate SBHCs and intends to identify the methods used by each SBHC in order to implement an oral health program. The study will also determine the best practices for implementing of oral health programs at Colorado’s SBHCs.

Dr. Bruce Dye
CareQuest Institute for Oral Health, $125,000
Facilitating Accessibility and Equity in Oral Health for those with Disabilities in the Rocky Mountain Region

The overall aim of this project proposal is to facilitate accessibility and equity in oral healthcare for those living with disabilities in Colorado and the broader Rocky Mountain region. We aim to (1) meaningfully engage with the disability community; (2) strengthen capacity and infrastructure by developing a safety net dental clinic in the Rocky Mountain region for those with disabilities and special healthcare needs (SHCNs); (3) work towards advancing state policy to reduce barriers to care; and (4) train the next generation of dental providers to recognize and prevent discrimination in providing dental care to many with disabilities.

 

March 2023

Drs. Trevor Williams and Hong Li were awarded a five-year grant of nearly $2.2 million from NIDCR entitled “In utero rescue of cleft lip and palate in a humanized mouse model”. Williams and Li have formed a collaboration with Dr. Jennifer Matsuda at National Jewish Hospital to explore the potential for treating pre-natal treatment to prevent common facial birth defects such as orofacial clefting.

Dr. Katherine Fantauzzo’s NIDCR funded project for over $2.35 million entitled “Srsf3-mediated alternative RNA splicing in craniofacial development” was recently awarded. The major goals of this project are to provide significant insight to mechanisms underlying gene expression regulation during mammalian craniofacial development and new therapeutic directions for the treatment of human craniofacial birth defects.

 

January 2023

Effective January 1, 2023, the salary limitation for Executive Level II is $212,100.

New FORMS-H Package for are now required for grant applications beginning January 25, 2023. 

A key change in FORMS-H is the implementation of the 2023 NIH Data Management Sharing Policy. The new policy requires all grant submissions to outline how scientific data and any accompanying metadata will be managed and shared, taking into account any potential restrictions and limitations. Additionally, costs associated with the implementation of the revised Data Management Sharing Plans can now itemized in grant budgets. December 2022

Dr. Tamanna Tiwari received a one-year award from the State of Colorado Department of Public Health and Environment (CDPHE) for $37,800. She will be collaborating with CDPHE’s Oral Health Unit in the evaluation of School-Based Health Centers (SBHC) in Colorado. The study seeks to identify best practices of SBHCs through the evaluation of common operations, successes and challenges SBHCs encounter during the implementation of oral health programs. The overall evaluation will help to better understand the needs for resources and assistance of Colorado’s oral health programs.

Data Management and Sharing Policy (DMS): Effective January 25, 2023, NIH submissions will require a DMS.

  • Grant submissions will need to outline how scientific data and any accompanying metadata will be managed and shared, taking into account any potential restrictions and limitations.
  • Compliance with awardee’s plan as approved by the NIH ICO.

DMS applies to all research funded or conducted in whole or in part by NIH that results in the generation of “scientific data.” Scientific data are defined as: “recorded factual material commonly accepted in the scientific community as of sufficient quality to validate and replicate research findings, regardless whether the data are used to support scholarly applications.”

 

October 2022

Dr. Tamanna Tiwari received a $115,000 supplement award for her current National Institute of Dental and Craniofacial Research R03 grant entitled “Assessing completeness and accuracy of the EHRs harbored at BigMouth Dental Data Repository.” The additional funding will focus on exploring new methods and strategies to improve the usability of Electronic Health Record (EHR) data for the purpose of oral health  research, dental education and patient care.

 

October 2022

Dr. Katherine Chin will be collaborating with Dr. Vanessa Fabrizio in the School of Medicine, Pediatrics. The new award is funded by Cincinnati Children’s Hospital Medical Center entitled “Reduction of bloodstream  infections from oral organisms in pediatric stem cell transplant: a randomized multicenter double-blind placebo-controlled study evaluating twice daily oral xylitol.” The study is being conducted at three institutions to evaluate the effectiveness of intraoral xylitol-wipe applications in reducing bloodstream infections among pediatric stem cell transplant patients.

Ms. Abigail Mumme-Monheit was awarded a Science Foundation Graduate Research Fellowships (NSF GRF). The NSF GRF four-year award of $184,000 provides a yearly stipend, tuition, benefits and fees. The purpose of the program is to broaden the participation in science and engineering of underrepresented groups, including women, minorities, persons with disabilities and veterans.
